Abstract: Depression and Alzheimer's disease (AD) are both prevalent diseases in older adults. Using the data sets from the Alzheimer's disease neuroimaging initiative (ADNI) study, the causal relationship between geriatric depression and AD is explored. The causal effect of geriatric depression is estimated on AD while controlling for ultrahigh-dimensional potential confounders, including DNA methylation, and finding that geriatric depression has a positive effect on AD. Moreover, a novel causal mediation analysis approach is developed to study the mediation effects of potential mediators on the causal relationship between geriatric depression and AD. Based on the real data analysis results, new prevention and treatment strategies are proposed for geriatric depression and AD by changing the selected confounders and mediators.
